Can I hook two radios at one time to the power supply, and useing only one radio at a time.

Yes you can as long as Both Radios do not pull the 32 Continuous Duty Amps.
I would guess you are talking about your Galaxy and the Yaesu FT-2800M.
With your setup you should have NO PROBLEM. 
The TRIPPLITE 40AMP POWER SUPPLY is Rated at: 32 Continuous Duty Amps.
